Nguyễn Đình Phúc xuất thân từ gia đình bình dân với người cha là giáo viên còn mẹ là tiểu thương. Từ nhỏ, ông tỏ ra say mê âm nhạc và hội họa nên đã học đàn cello. Nguyễn Đình Phúc còn là sinh viên dự bị của Trường Mỹ thuật Đông Dương khóa 1941 – 1942. Khởi đầu sự nghiệp, ông biểu diễn cello cho một gánh xiếc lưu động. Trong thời kì tân nhạc Việt Nam dần hình thành, ông là một nghệ sĩ độc lập. Thời gian này, Nguyễn Đình Phúc đã có sáng tác đầu tay là bài "Lệ thu", nhưng từ bài "Cô lái đò" phổ thơ Nguyễn Bính mới giúp ông có một vị trí trong dòng nhạc lãng mạn Việt Nam trước Cách mạng Tháng Tám.
